Neto do rei Vítor Manuel II, rei da Sardenha-Piemonte (entre 1859 e 1861) e da Itália (1861-1878), ascendeu ao trono italiano após o assassinato do pai, Humberto I (rei de 1878 a 1900). Numa primeira fase, desempenhou unicamente funções constitucionais, mas, durante a crise que se seguiu à Primeira Guerra Mundial, teve um importante papel nas decisões políticas. Em 1922, encarregou Benito Mussolini, depois da sua marcha sobre Roma, da formação do novo governo e, nos anos seguintes, apoiou plenamente o regime fascista. Em conseqüência da política expansionista de Mussolini, de 1936 a 1941 e de 1939 a 1943, respectivamente, foi igualmente imperador da Abissínia e rei da Albânia. Após a derrota militar da Itália na Segunda Guerra Mundial, participou em 1943 na destituição de Mussolini por parte do Grande Conselho Fascista e assumiu o poder. Em 1944, renunciou a favor de seu filho Humberto II e, em 1946, abdicou e exilou-se. Pouco depois, foi proclamada a República.
